So I've softmodded my 4.3U Wii, now what?

I followed the guide for softmodding my 4.3U Wii (completely new to the scene here), everything went as instructed, no problems at all. I have the apps at the end of the tutorial on the SD card in the Wii, and the four new channels appear as the guide shows them.

Now I have no idea how to play my backups. I burned one ISO to a disk, figuring I could use it in the Backup Launcher Channel. I used ImgBurn, and burned at a very slow speed (3x I think). Disc won't read; I tried multiple copies with no luck.

I tried hooking my external HDD to the Wii and using the Configurable USB Loader that had the same ISO on it. The HDD is plugged into the outermost slot as instructed. If the hard drive is connected when I boot the CFG USB Loader channel, it will freeze at the first/splash screen. If the HDD is disconnected, I can get past that screen to the screen with the Wiihacks.com logo, and it begins the 30-second count for detection. I can plug my HDD in then, and it will stop the countdown and say "Ok!" but then will freeze. In both cases I have to reset the Wii.

I'm completely new to this and would much appreciate someone instructing me on how to get my backups to work. Thanks!

How new is your Wii? If it's a little over a year old or newer, it won't play optical backups. On the USB HD issue: did you check to see if the device was on the compatibility list?

Ah, maybe. The hard drive I was trying was ripped out of an old computer and is using a USB adapter. That could be a huge problem.

As for whether or not it will support optical media, I was told to look at the serial number on the bottom of the Wii, and if it started with LU64 or above, that it wouldn't support DVD backups. Mine starts with LU22 or LU23, can't remember, so maybe I'm ok on that.

That's not exactly scientific, nor necessarily even true. Go here and enter the full serial number, see what result you get.

Alright, thanks for the link. Both of the models it says I may have have a big "N/A" for the DVD Board column. I guess that means I'm out of luck. Correct me if I'm wrong, but aren't Wii games on standard DVDs? Why is there no support?

I have a compatible external HDD I can use, but, can the CFG USB Loader channel detect an NTFS drive? If I format to FAT32, the ISOs can't copy over, and the tutorials for splitting the files to fit on FAT32 seem a little tough.

Your question doesn't even make sense. Standard DVDs? Sure; standard readily-readable format for say a PC, no. We do have guides on formatting (whether NTFS, FAT32, WBFS) in the Recommended Guides section.
