Of course had to try this!

Extracting, editing, replacing and renaming all went well until I tried playing:

After pressing (2) at the "hold your Wiimote sideways to play", I just get a black screen and the Wii hangs.

And that's after using the Partition Builder app right?

Did you happen to use or move around any of the files included in the "Reggie Extras" folder?

I just edited level 01-01.arc and replaced that with WiiScrubber. I did not use Partition Builder. I was under the impression I needed Partition builder only if there were lots of changed files. Will give it a go.

No you don't need to use it, you can, but you don't have to.

If it's black screening after editing a level it usually means a messed up entrance - like the point mario first starts at. Here's a good post on that:

the last option on the palette looks like a door, click on that, then right click on the stage where you want mario to start, and dont change anything, it should be all 0's in the option box that appears for it.. and thats how you set up the starting entrance.. with "enterable" checkmarked. and the normal option which should already be selected.

make sure you have a good amount of space behind marios starting position or he could be popping into your screen at the very edge and it looks really bad that way..
But just to cover our bases, make sure the original ISO works fine. Even if it did before, humor me and try it again. Then replace the custom level once more so we can be absolutely sure it's the added file causing a black screen.

Im not sure, Its just when I'm finished and I put the ISO back together with the Partition Builder (either version), I just get a black screen. And I kind of figured out Reggie a little more.. I wasn't even looking at the "Layers" at first.

Where is the black screen happening specifically? Also skim over the last few posts as you and own1970 are having similar issues.

Just for kicks I tested rebuilding my partition with v1.2 a couple hours ago and the same level included with this guide again to be sure, and it does work. I'll help you both as much as I can but I do need to know more info.

Covering the bases...good thought, must be somewhere in the chain of tooling:

    Here is what I did:
Using Wii Backup Manager 0.3.4 I copied the NSMB file to my PC as .wbfs, then using the same program converted to .iso (could have been done in one go)
Using WiiScrubber extracted everything to "Original Mario Contents"
    Copied everything to "Newer Mario"
    Edited 01-01.arc
Replaced 01-01.arc with edited version using WiiScrubber
    Renamed and Re-ID'd with WIIISORenamerv3
Copied the Newer.iso to USB HDD
===> Black screen after "hold wii remote sideways"

After your comments, I replaced 01-01.arc with the original 01-01.arc using WiiScrubber and copied it again to de USB HDD
===> Black screen after "hold wii remote sideways"

So, the level should be OK, I must be doing something else wrong. Will try again tomorrow starting all over. Keep you posted and am open for suggestions :-)
