There are two ways of backing-up games.

I'll be assuming that you have cIOS installed and that you have and know what a WBFS partition is.
a) Most USB loaders have an "Install Game" function. This allows the Wii to read Wii game discs and copy the content to your drive.
If you are using USB Loader GX, right next to the Setting buttion, you should see a plus sign. Select that, and install the game.
If you are using Configurable USB Loader/(Waninkinko's) USB Loader, you could press the + button on your Wii remote.
b) If you want to do this on Windows, you will need:
Stick your Wii disc inside the DVD drive.
Start up ImgBurn. and select "Create image file from disc."
Save it somewhere that you will remember, and is at least 5GB (Most games would usually cost you 4.7 or 4.8 GB of free space).
You could stop here if you only wanted an ISO file.
Continue, if you want it to be played on USB loaders.
After creating the ISO file, start up WBFS Manager
On the top corner where it says Drive, select your WBFS drive.
On the bottom, click browse, and open the new ISO file.
Then select add to drive.
